PURPOSE: A mutant hepatitis C virus(HCV) containing a reporter gene is provided to research infection using alive cells and study virus assembly and release. CONSTITUTION: A polynucleotide containing a mutant HCV genome has JFH1 strain genome RNA E2 region or p7 region in which one or more base sequences are substituted. One or more bases among 2027th-2029th bases of sequence number 1 in E2 region are substituted with other base. The substituted 2027th-2029th bases do not encode threonine. One or more bases among 2027th-2029th bases of p7 region are substituted with other base. The substituted 2027th-2029th bases do not encode arparagine. A method for screening anti-HVC agent comprises: a step of introducing a polynucleotide containing the mutant HCV genome to a host cell under the presence of test sample and culturing the cell; and a step of evaluating anti-HCV effect of the test sample.
