Provide is a method for purification of recombinant proteins using a self-cleaving protein intein, which comprises the steps of: recombinantly expressing a fusion protein which comprises a target protein domain, a self-cleaving intein and one or more domains for binding hydrophobic granules in host cells, wherein the self-cleaving intein is located between the target protein domain and one or more domains for binding hydrophobic granules; making the fusion protein releasing from host cells, then adding hydrophobic granules to it and incubating them; collecting hydrophobic granules after incubation and adding lysis buffer to allow the self-cleaving intein of the fusion protein to self-cleave; separating and removing the hydrophobic granules to obtain basically purified the target protein solution. A kit useful for the method is also provided.
