<P>PROBLEM TO BE SOLVED: To provide a method for cloning immunoglobulin variable domain (IGVD) sequences derived from immunoglobulin, and to provide a repertory library of the immunoglobulin variable domain sequences provided by the method. <P>SOLUTION: Provided is the expression library comprising the IGVD polynucleotide sequence and the repertory of the sequence, capable of being obtained by the method which includes synthesizing a first strand cDNA from mRNA of a sample by using universal primer, producing double strand DNA by synthesizing a second strand DNA by using species-specific primer, cleaving the double strand DNA with a restriction enzyme specific for a naturally generating restriction site and a restriction enzyme specific to restriction site encoded by the species-specific primer, and cloning the obtained variable domain fragment sequence into a vector. <P>COPYRIGHT: (C)2009,JPO&INPIT |
