A natural composite amino acid seasoning containing highly concentrated glutamate and a preparation method thereof are provided to replace a conventional fermenting method and to improve a taste. A method for preparing a natural composite amino acid seasoning comprises the following steps of: hydrolyzing proteins representing a vegetable protein selected from the group consisting of defatted soybeans, cone gluten and wheat gluten or an animal protein or a fish protein and alkalizing them with caustic soda; concentrating them in an alkali state of pH 8.0 ~ 11.0 to prepare a concentrated amino acid liquid; adding the concentrated amino acid liquid to hydrochloric acid and adjusting to an isoelectric state of pH 2~5 in order to induce precipitate and neutralize it and obtaining a liquid or paste phase; adjusting a corn gluten meal decomposing liquid T-N to pH 9.5~10.5 by using 48% caustic soda; standing as it is at 75°C~85°C for 2-10 hours and concentrating the corn gluten meal decomposing liquid T-N to 4% or greater; adjusting to pH 3.2 by using concentrated hydrochloric acid and educing the crystallization of glutamic acid hydrochloride; and growing the crystallization and neutralizing with 40% caustic soda.
