摘要 The subject matter described herein relates to a process for preparing an antibody, in one embodiment a Fab', in very good yields and purity, under conditions where the presence of heavy and light chain antibody fragments is minimized. More particularly, the subject matter described herein relates to a process for reducing F(ab)2 to primarily heavy and light chains, followed by a reoxidation step that is selective for making Fab' in very good yields and purity by reforming the disulfide bridge between the heavy and light chains. The reoxidation step is carried out to minimize the presence of heavy and light chain, minimize the generation of F(ab')2 and maximize Fab'. In one embodiment, the subject matter described herein relates to a process for preparing an antibody composition, in one embodiment a Fab' liposome composition, having specific binding activity for alpha-V-integrin receptors. The composition is intended for use in treating conditions characterized by cells that express any alpha-V-comprising integrin, such as alphavbeta3, alphavbeta5, and alphavbeta6 receptors.
