PROBLEM TO BE SOLVED: To provide an atmosphere-regulating agent with excellent handleability, and to provide a cell culture method and a cell culture kit using the agent. SOLUTION: The cell culture kit 1 has a dish 12 for a medium and a dish 13 for a capsule stored in a container body 10, and the inside thereof can be hermetically sealed by a lid 11 for sealing. The medium 12a for cells and the cells are set on the dish 12 for the medium. At the start of the culture, water 13a is poured to the dish 13 for the capsule so that an atmosphere-regulating capsule 14 can be soaked therein. The atmosphere-regulating capsule 14 includes an agent 14b containing a carbonate and acids encapsulated with a water-soluble encapsulating body 14a. When the atmosphere-regulating capsule 14 is soaked in the water, the encapsulating body 14a is dissolved after a prescribed time period, and the agent 14b is dissolved in the water 13a to cause chemical reaction to generate carbon dioxide gas.
