#CMT# #/CMT# Compositions (I) containing (a) a unicellular alga of the genus Asterionella, (b) caffeine and (c) theobromine. #CMT# : #/CMT# Independent claims are also included for (1) a method (M1) for the production of (I) by centrifuging a culture of the algae, removing the supernatant and mixing the algal paste with the other components (2) a method (M2) for the production of (I) as in M1, except that the algal paste is resuspended and the suspension is mixed with the other components (3) a method (M3) for the production of (I) by making algal paste as in M1, splitting the algal cells, removing the membrane debris by a suitable method and mixing the cellular extract with the other components (4) cosmetic formulations containing (I) and a cosmetically-acceptable vehicle . #CMT#USE : #/CMT# For the preparation of cosmetic formulations, preferably containing 0.1-100 wt.% of the composition (I) (claimed). Also claimed is the use of these cosmetic formulations for topical application for slimming. #CMT#ADVANTAGE : #/CMT# The components of this composition show a synergistic action in the inhibition of phosphodiesterase (an enzyme involved in lipid metabolism), resulting in an increase in intracellular AMPc and therefore a lipolytic effect. #CMT#ORGANIC CHEMISTRY : #/CMT# Preferred Compositions: Compositions (I) containing 105>-101>0> algal cells/ml (or equivalent biomass), 0.2-4 wt.% caffeine and 0.005-5 wt.% theobromine. Preferred Formulations: Cosmetic formulations containing 104>-108> algal cells/ml, in the form of lotions, emulsions, creams, gels or patches. #CMT#EXAMPLE : #/CMT# When tested for its effect on the activity of phosphodiesterase (PDE; human adipose tissue extract) in the hydrolysis of AMPc, pure caffeine (in 50-mM TRIS buffer with 5-mM MgCl2; pH 7.5) showed an IC50 value of 0.45-mM (0.0086 vol.%) and pure theobromine showed an IC 50 value of 0.0115 vol.%. An extract of Asterionella with 400 x 106> cells/ml in its culture supernatant (0% theobromine) and 20% ethanol (made by freeze drying the centrifuged culture and resuspending in ethanol) showed an IC50 of 0.17 vol.%. A similar extract with 0.02% theobromine, 20% ethanol, modified by the addition of 2 wt.% caffeine and 2 wt.% extra theobromine, showed an IC50 of 0.0014 vol.%, i.e. an activity 6 times greater than that of caffeine and 4 times that of theobromine. A lotion according to the invention contained 0.1 wt.% benzophenone-9, 6 wt.% methylpropanediol, 0.2 wt.% disodium EDTA, 2 wt.% Acacia, 0.6 wt.% xanthan gum, 0.1 wt.% allantoin, 0.3 wt.% lysine (50%), 10 wt.% extract of Asterionella (107> cells/ml; ethanol 30%; caffeine 2%; theobromine 2%), 0.8 wt.% octyldodeceth-30, 0.15 wt.% perfume, 0.2 wt.% potassium sorbate and sea water (q.s. 100). |
