PROBLEM TO BE SOLVED: To provide a method for preparing a polypeptide modified with a water soluble polymer. SOLUTION: The method for preparing a polypeptide modified with a water soluble polymer comprises the step of causing a water soluble polymer having an aldehyde group to react with a polypeptide in the presence of a reductant selected from the group consisting of 2-picolineborane and 3-picolineborane. A method according to the above method wherein the water soluble polymer segment of the water soluble polymer having an aldehyde group is selected from the group consisting of dextran, poly(N-vinylpyrrolidone)polyethyleneglycol, a propyleneglycol homopolymer, a propyleneoxide/ethyleneoixde copolymer, a polyoxyethylated polyol and polyvinyl alcohol is also disclosed.
