摘要 The present disclosure relates to methods of characterizing protein-prote in interaction or conducting immunoassays in a biological sample using self- assembled monolayers (SAMs) and matrix-assisted laser desorption / ionizati on time of flight mass spectrometry (SAMDI). The biological sample may be ob tained from a living subject, such as a human or animal clinical sample cont aining multiple unknown proteins and/or antigens. Label-free methods of iden tifying protein-protein interactions, antigen-antibody binding and/or diagno sing a medical condition based on analysis of a biological sample using SAMD I are also provided, as well as biochips comprising surface bound proteins a nd/or antibodies and methods of making these biochips. The methods and bioch ips are useful, for example, for identifying protein-protein binding interac tions and/or conducting immunoassays in samples such as humoral fluids or ot her clinical samples, cell lysates, tissue lysates, tumor lysates, and sampl es obtained, isolated or derived from animals or plants.
