Disclosed is an in vitro method of selecting a cell producing a secreted polypeptide, the method comprising: providing a cell population, wherein the cell population comprises a cell comprising a heterologous nucleic acid encoding a secreted polypeptide; contacting the cell population with an antibody that specifically binds to the secreted polypeptide; detecting the binding of the antibody to the secreted polypeptide while transiently associated with the plasma membrane during protein secretion, wherein the detecting occurs without adding a substance to the cell that forms a matrix around the cell or immobilizes the secreted polypeptide near the cell; and selecting the cell based upon the presence or amount of the antibody bound to the secreted polypeptide transiently associated with the plasma membrane during protein secretion. |
