METHOD FOR PRODUCING EXTRACT OF HORSE CHESTNUT LEAF

A method for producing the extract of horse chestnut leaf is provided to improve MMP(matrix-metalloproteinases)-inhibitory effects of the horse chestnut leaf extract, so that the horse chestnut leaf extract is useful for prevention and treatment of MMP-mediated disease including periodontal disease. A method for producing the extract of horse chestnut leaf comprises the steps of: (1) drying and pulverizing horse chestnut leaves; (1-1) optionally subjecting the pulverized horse chestnut leaves to supercritical treatment using carbon dioxide as fluid at 25-45 deg. C and 200-350 atmospheric pressure (2) heating the pulverized horse chestnut leaves in 25% alcohol solution to 50-65 deg. C; (3) filtering the extracted solution; (4) concentrating the filtered solution, and (4-1) optionally fractionating the concentrated extract of horse chestnut leaf with hexane, ethylacetate, butanol and water, sequentially. A composition for prevention and treatment of MMP-mediated disease comprises the extract of horse chestnut leaf. Further, the alcohol is methanol, ethanol or a mixture thereof.
