A composition comprising orotic acid or a derivative thereof is provided to enhance biosynthesis of hyaluronic acid and/or glycosaminoglycan in the cartilage cells, synovial cells, myofibroblasts, etc, and to prevent or treat arthritis, prolapsed intervertebral disk, winkles, dry eye syndrome, atopic dermatitis, etc. A composition for enhancing biosynthesis of hyaluronic acid and/or glycosaminoglycan comprises at least one selected from orotic acid and derivatives thereof as an active ingredient. The orotic acid derivative is at least one selected from the group consisting of orotates of sodium, potassium, calcium, magnesium, zinc, manganese, selenium, chrome, lithium, cobalt, iron and ammonium, choline orotate, creatine orotate, carnitine orotate, arginine orotate, lysine orotate, chitosan orotate, chitosan oligo orotate, polylysine orotate, orotidylate(orotidine monophosphate), orotidine and dihydroorotic acid.
