A method for assessing the phagocytic activity of neutrophils comprises the freeze-drying of the fresh yeasts Saccharomyces cerevisiae at the temperature of 100 °С, the incubation of the yeasts in the boiling water for 60 min, the washing in the buffer solution adjusting pH to 7.2-7.4, the five-fold dilution with 0.9% solution of NaCl. Then the specified amount of the diluted yeasts upon the treatment above is added to the sample of the peripheral blood. In addition, to the blood sample of 3 mL, 0.2 mL of heparin and 0.05 mL of yeast suspension is added successively. The mixture is incubated at the temperature of 37 °С shaking it every 5 min. Prior to the preparation of the smears, the mixture is cooled in water and centrifuged at 1500 rpm for 5-7 min. The supernatant is removed and the pellet is resuspended. The sample of 0.2 mL is taken from the rest of the whole sample, and the ratio of yeasts : phagocytes is adjusted to 10:1. |
