PROBLEM TO BE SOLVED: To provide a method for measuring the cholesterol level in low-density lipoprotein (LDL), in a biological specimen, such as sera and plasmas, and to provide a reagent for the measurement. SOLUTION: The method for measuring the cholesterol level in low-density lipoprotein by an enzymatic reaction comprises a first reaction for preferentially eliminating high-density lipoprotein and ultra-low-density lipoprotein in the specimen and cholesterol in chylomicrons, in a buffer solution of pH of 7.6-9.0 containing cholesterol esterase, cholesterol oxidase and a substance for inhibiting reactivity of these enzymes to a low-density lipoprotein, and a second reaction for adding a substance to make the inhibition action of both the enzymes reduce or eliminate in the first reaction, with respect to the cholesterol in the low-density lipoprotein to the reaction liquid.
