PROBLEM TO BE SOLVED: To provide a method for measuring the content of methylated DNA:X. SOLUTION: The method for measuring the content of methylated DNA:X comprises the following procedure: A single-stranded DNA:A and a single-stranded immobilized oligonucleotide:B are bound to each other from a specimen to select A. With A as a template, B as a primer:PR is extended once and formed as a double-strand:C, which is then subjected to digestion treatment with a methylation-sensitive restriction enzyme followed by removing the free digested product formed, wherein as the prior step, C as an undigested product is isolated once in a single-stranded state, and the single-stranded DNA(positive strand):D formed and B are bound to each other to select a single-stranded DNA:E. With the selected E as a template, B as a primer PR is extended once and formed as a double-strand. With a single-stranded DNA(negative strand):F as a template, G as a specific PR:G is extended once to form F as a double-strand. Further, the thus extendedly formed double-stranded DNA by the above operation is separated into a single-stranded state once. Subsequently, the above operation is repeated to amplify/quantify X.
