A culture medium composition for culturing human mesenchymal stem cells is provided to secure bio-stability by using minimum amount of human serum. A method for selecting and culturing the human mesenchymal stem cells is provided to express a marker of CD9 increasing proliferation capability and adherence capability. A culture medium composition for culturing cells including human serum without animal serum comprises 50-60 vol.% of DMEM(Dulbecco's Modified Eagle's Medium), 40-50 vol.% of IMDM(Iscove's Modified Dulbecco's Medium), 4-6 mug/ml of insulin, 4-6 mug/ml of transferrin, 4-6 mug/ml of selenium, 10^-7-10^-9M of dexamethasone, 80-120U penicillin/800-1,200 U streptomycin, 5-15 ng/ml of FGF, and 1-10% of the human serum. A method for selecting and culturing human mesenchymal stem cells with increased proliferation capability and adherence capability comprises the steps of: (a) culturing human mesenchymal stem cells derived from an adipose tissue or bone marrow using the culture medium composition; and (b) isolating stem cells expressing a CD9 marker from the culture material obtained from the step(a). Further, the isolating step is performed by isolating cell having CD9 antigen using a fluorescent activated cell sorting system or a magnetic activated cell sorting system. |
