The present invention provides processes for the preparation of substituted 2-(2,6- dioxopiperidin-3-yl)isoindole-l,3-diones which are useful, for example, for preventing or treating diseases or conditions related to an abnormally high level or activity of TNF-a.The invention can provide cost-effective and efficient processes for the commercial production of substituted 2-(2,6-dioxopiperidin-3-yl)isoindole-l,3-diones, including, but not limited to, 4- [(N,N-dimethylhydrazono)methyl]-2-(2,6-dioxopiperidin-3-yl)isoindole-1.3-dione, 4-aminornethyl-2-(2,6-dioxopiperidin-3-yl)isoindole-l,3-dione, and 4-[(acylamino)methyl]-2- (2,6-dioxopiperidin-3-yl)isoindole-l,3-diones.
