The present invention relates to a process for making the improved C-terminal Histidine tagged HOXB4 recombinant protein (TAT-HOXB4H) which containing 6 Histidine residues in the C-terminus. The generation of the C-terminal Histidine tag increases the amount of TAT-HOXB4H recombinant protein to be purified about 3-4 fold compared to the original TAT-HOXB4. The present invention also relates to a construct for preparing the TAT-HOXB4H recombinant protein with increased yield to facilitate expansion of stem cells.
