PROBLEM TO BE SOLVED: To provide DNA segments encoding the recognition and cleavage domains of an FokI restriction endonuclease.

SOLUTION: The 41 kDa N-terminal fragment constitutes a recognition domain of the FokI restriction endonuclease while the 41 kDa C-terminal fragment constitutes a nuclease domain of the FokI restriction endonuclease. The present invention relates to hybrid restriction enzymes (Ubx-FN) comprising the nuclease domain of the FokI restriction endonuclease by linking ultrabithorax (Ubx) homeo domain to the cleavage domain (FN) of the FokI. Additionally, the present invention relates to the construction of two insertion mutants of FokI endonuclease.
