<P>PROBLEM TO BE SOLVED: To provide a method and a kit, etc. for releasing the transcriptional regulation caused by a repeated sequence in a gene, and establish a system capable of producing a protein in a large amount. <P>SOLUTION: The method of the present invention contains a step to improve the promoter activity inducing the expression of a gene encoding the target protein. The method may further contain one or more steps comprising: (a) a step to culture and select the cell having undergone gene amplification in a medium containing a gradually increasing agent; (b) a step to improve the promoter activity to induce the expression of a gene encoding the target protein; (c) a step to lower the methylation level of DNA by treating the cell having undergone gene amplification with 5-aza-2'-deoxycytidine; and (d) a step to select the above mammalian cells having undergone gene amplification on double minute chromosomes. <P>COPYRIGHT: (C)2008,JPO&INPIT |
