<P>PROBLEM TO BE SOLVED: To provide a new DNA-cleaving enzyme protein and to provide a gene thereof. <P>SOLUTION: The new DNA-cleaving enzyme is derived from Pyrococcus furiosus, Pyrococcus abyssi, Pyrococcus horikoshii or Thermococcus kodakaraensis. Since the enzyme recognizes and degrades the 3'-terminus of a single-stranded DNA, the enzyme can be utilized for technical development for cleaving an optional DNA strand in a specific site by utilizing a difference between single strands and double strands of the DNA in structure. Furthermore, since actions can be exhibited at high temperatures, the enzyme can be used in such a state that the cleavage reaction resistance by formation of a high-order structure of the DNA strand itself which is a substrate is avoided. <P>COPYRIGHT: (C)2008,JPO&INPIT |
