PROBLEM TO BE SOLVED: To provide an industrial method for production by which auto-hydrolysis causing the deterioration of optical purity of a substrate (aβ-amino acid ester) readily hydrolyzable with water can nearly completely be suppressed and an optically active N-substituted-β-amino acid can completely be recovered. SOLUTION: Water is selectively reacted with only one of enantiomers of an N-substituted-β-amino acid alkyl ester (a racemic mixture) represented by general formula (I) (wherein, P represents a protective group; R<SP>1</SP>represents an alkyl group, an alkenyl group, a cycloalkyl group, an aralkyl group or an aryl group; and R represents an alkyl group) in the presence of a hydrolase in an organic solvent to produce an optically active (R or S)-N-substituted-β-amino acid.
