A method of preparing silica nanopowder comprised of spherical single particles by flame spray pyrolysis adopting a two-fluid nozzle is provided to prepare spherical silica nanopowder by directly oxidizing droplets sprayed at a high pressure from a silicon alkoxide solution in a flame using the flame spray pyrolysis. A method of preparing silica nanopowder comprised of spherical single particles by flame spray pyrolysis adopting a two-fluid nozzle comprises: a droplet spray step(S100) of spraying silicon alkoxide droplets as a liquid phase silicon compound through a two-fluid nozzle type droplet spray of a flame reactor; a flame producing step(S110) of simultaneously flowing inert gas, hydrogen, oxygen and air into a burner of the flame reactor to produce a flame; a nanopowder producing step(S120) of passing the liquid phase silicon compound through the flame of the burner to produce silica nanopowder with an average particle size of 18 to 68 nm; and a recovery step(S130) of collecting the produced nanopowder through a particle collection part to recover the nanopowder, wherein the silicon alkoxide is tetraethylorthosilicate or tetramethylorthosilicate.
