A composition for culturing plant tissue comprising luminous or fluorescent materials is provided to exhibit various colors from layers of the medium, and inhibit the layer boundary of the medium from becoming dim while growth of plant tissue is not inhibited, so that a multiple layer medium for culturing plant tissue having clearly different colors is produced. The composition for culturing plant tissue comprises 0.1-0.5% of luminous or fluorescent materials, 0.5-1.3% of colloidal materials, 0.1-1% of inorganic nutrition elements, 2.0-5.0% of carbon source, 0.001-0.005% of vitamins and a remaining amount of water, wherein the luminous material is zinc sulfate(ZnS:Cu); the fluorescent material is zinc sulfate or cadmium zinc sulfate; the colloidal material is agar, agarose or gelite; the inorganic nutrition elements include MgSO4, MnSO4, CuSO4, NH4NO3, CaCl2, KI, KH2PO4, Na2MoO4.2H2O and FeEDTA(ethylene diamine tetraacetate); and the carbon source is glucose, fructose, mannose, ribose, xylose, sucrose, melibiose, cellobios, lactose, amylase, starch, raffinose, sorbitol, mannitol or glycerol. The multiple layer medium for culturing plant tissue is prepared by pouring and solidifying the composition by turns.
