PROBLEM TO BE SOLVED: To enable long-term stable operation by inhibiting a deterioration in membrane flux of an RO device for carrying out desalting and/or removal of organic matter. SOLUTION: In an operation method of a reverse osmosis membrane separator for carrying out desalting and/or removal of organic matter, water to be treated whose pH has been adjusted to 9.5-13 by addition of an alkali is used as flushing water to temporarily flush the reverse osmosis membrane separator. By carrying out flushing with the alkaline water to be treated during operation, a membrane surface is washed effectively to inhibit the deterioration of the membrane flux, which enables a stable operation for the long term. Almost all of supply water does not permeate a membrane during the flushing, therefore concentration on the membrane surface does not occur, which prevents the problems of scale generation on the membrane surface even when the supply water is alkaline.
