发明名称 High Efficiency Micro Reaction Block and Manufacturing Method Thereof
摘要 A method for preparing a micro reaction block is provided to be able to obtain the accurate reaction result even with input of an extremely small amount of a sample by attaching enzyme to a carbon nanotube. The method comprises the steps of: (a) after obtaining carbon nanotube compact body from carbon nanotube powder, putting it in a falcon tube and adjusting the pH thereof using sodium hydroxide with decomposing it through an ultrasonic cleaner; (b) after filtering the product obtained from the step(a) through a nylon membrane filter, raking 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide out from the membrane filter and then reacting it with n-hydroxysuccinimide at room temperature; (c) after taking out supernatant not-reacted in a centrifugal separator using a pipette and washing it with phosphate buffer sharing(PBS), repeating from the decomposition using the ultrasonic cleaner to the washing step with the PBS 2-3 times; (d) after reacting horseradish peroxidase(HRP) with glucose oxidase(GOX) and taking out supernatant from the centrifugal separator using a pipette, repetitively washing it with the PBS and putting 2-2'-azino-bis(3-ethylbenzothiazolinee-6-sulfonic acid)diammonium salt and hydrogen peroxide therein to identify the HRP by the color change; (e) obtaining a mixture where the enzyme is attached to the sticky carbon nanotube compact body; (f) filling the mixture in a groove portion of a mold; (g) applying heat and pressure to the mixture with covering a cover thereon to mold and harden the carbon nanotube mixture; ad (h) after taking the hardened carbon nanotube out of the mold, molding it into a desired shape to obtain a micro reaction block where the enzyme is attached to the carbon nanotube.
