PROBLEM TO BE SOLVED: To provide a method for practically efficiently obtaining an isobutyric acid ester body of a glycerol acetonide having high optical purity by utilizing an easily prepared or easily available enzyme. SOLUTION: The method for producing the optically active glycerol acetonide isobutyric acid ester comprises reacting an ester hydrolytic enzyme derived from the liver of a rabbit, or an ester hydrolytic enzyme derived from microorganisms belonging to the genuses of Serratia, Alcaligenes, Burkholderia and Enterobacter with a racemic body of the glycerol acetonide isobutyric acid ester in an aqueous solvent to hydrolyze the racemic body of the compound stereoselectively, and collecting the remaining optically active glycerol acetonide isobutyric acid ester.
