A process for production of a protein having a non-natural amino acid integrated therein, the process comprising expressing in an eukaryotic cell an aminoacyl tRNA synthetase, a nucleic acid which comprises a sequence having a tRNA nucleotide sequence derived from an eukaryotic cell ligated to the 5' terminus of a tRNA nucleotide sequence capable of binding to a non-natural amino acid in the presence of the aminoacyl tRNA synthetase, a non-natural amino acid, and a gene for a desired protein having a nonsense mutation at a predetermined position to integrate the non-natural amino acid at the non-sense mutation position in the protein, thereby expressing a protein having a non-natural amino acid integrated therein.
