A call processing method based on number portability is provided to enable a calling party's exchange to directly query location information to an NPDB and acquire the location information of a receiving mobile communication terminal even when an error occurs in an HLR, having an initial address, during a call processing process based on the number portability using a QoR(Query on Release) process, thereby improving a call processing rate and improving QoS(Quality of Service). A call processing method based on number portability comprises the following steps of: enabling an exchange of a mobile communication system, which a phone number of a receiving mobile communication terminal indicates, to request location information of the receiving mobile communication terminal from an HLR(Home Location Register)(S303); notifying a calling party's exchange of an error of the HLR if the location information of the receiving mobile communication terminal is not received from the HLR within a designated time(S309); enabling the calling party's exchange to query the location information to an NPDB(Number Portability DataBase) and receive a response(S311,S313); and enabling the calling party's exchange to set a call to a real receiving exchange(S315).
