Recombinant DNA (I) derived from the genome of a salmonid alphavirus (SAV) is new. It comprises (a) a transcriptional promoter and, downstream of this and under its control, (b) a spacer of at least 5 nucleotides (nt) and (c) the cDNA (Ia) from the genomic RNA of an SAV. Independent claims are included for: (1) preparing an RNA replicon of SAV by introducing (I) into a host cell and culturing the cell; (2) the RNA replicon prepared by method (1); (3) method for preparing a recombinant SAV by introducing (I), or the replicon of (2), into a host cell where all structural proteins required for encapsidation are expressed, then growing the cells; (4) recombinant SAV prepared by method (3); and (5) vaccine that contains (I), the replicon of (2) or recombinant SAV of (4).
