The present invention relates to recombinant DNA which encodes the <Italic>Nhe</Italic>l restriction endonuclease as well as <Italic>Nhe</Italic>I methyltransferase; expression of <Italic>Nhe</Italic>l restriction endonuclease from <Italic>E. coli</Italic> cells containing the recombinant DNA.
