Provided is a method for preparing N(5)-ethylglutamine(theanine) in which amidation is effected under same condition as that of deprotection reaction under medium state that phthaloyl group does not left completely, and thus amidation and deprotection reactions are simultaneously effected in one reaction. The method for preparing a theanine comprises the steps of (i) reacting L-glutamic acid derivative protected by phthaloyl group represented by the formula 1 and ethylamine to sequentially effect an amidation and a deprotection under same condition, and (ii) adding an appropriate organic solvent to the liquid reactant the step(i), and extracting and filtering a theanine of the formula 2 to obtain the theanine. In the formula 1, R is a C1-C5 alkyl group or benzyl group, preferably methyl or ethyl group, each of X2, X2, X3, and X4 is independently hydrogen or halogen atom, or nitro group, preferably hydrogen atom.
