摘要 A system for functional analysis of polypeptides is provided to be able to directly culture yeast cells and mammalian cells at the same time and determine function of polypeptide having ligand activity without purification of the polypeptide from the yeast cells producing the heterologous polypeptide. The system for functional analysis of polypeptides comprises: a non-mammalian cell culture which is cultured in a non-mammalian cell culture medium and expresses a heterologous polypeptide that is either displayed on its cell surface such that the polypeptide is the predominant polypeptide displayed on the cell surface or the polypeptide is secreted; a mammalian cell which is cultured in a mammalian cell culture medium and has a reporter construct capable of being specifically bound to the heterologous polypeptide; and a tool for detecting interaction between the heterologous polypeptide expressed in the non-mammalian cell and the reporter construct expressed in the mammalian cell. And the system analyses the function of the heterologous polypeptide by detecting the interaction between the heterologous polypeptide expressed in the non-mammalian cell and the reporter construct expressed in the mammalian cell. In the system, the non-mammalian cell culture medium is not suitable for culturing mammalian cell, and the mammalian cell culture medium is suitable for culturing mammalian and non-mammalian cell.
