METHOD FOR OVERPRODUCING A SPECIFIC RECOMBINANT PROTEIN WITH P. CINNABARINUS MONOKARYOTIC STRAINS

Method for preparing a recombinant protein (I) by overexpressing the appropriate gene (II) in a monokaryotic strain (A) of a filamentous fungus of the genus Pycnoporus (Basidiomycetes). Method for preparing a recombinant protein (I) by overexpressing the appropriate gene (II) in a monokaryotic strain (A) of a filamentous fungus of the genus Pycnoporus (Basidiomycetes) comprises: (A) culturing (A) that has been transformed with an expression vector containing (II), under control of a promoter, endogenous or exogenous, constitutive or inducible; (B) inducing the promoter, if required; and (C) recovering, and optionally purifying, (I) produced in the culture medium. Independent claims are also included for the following: (1) expression vector, e.g. pELP, that includes a 2527 bp sequence (S3), i.e. the pLac promoter of the laccase gene endogenous to P. cinnabarinus, or its functional derivatives formed by substitution, addition or deletion of one or more nucleotides; and (2) host cells transformed with the vector of (1).
