METHOD FOR RECONSTRUCTION OF BLOOD CELL FROM GENE-TRANSFERRED HEMATOPOIETIC CELL
摘要

[PROBLEMS] To provide a method for producing a hematopoietic cell engraftment composition comprising a gene-transferred hematopoietic cell by using a chromosomally unintegrated viral vector, and to provide a method for reconstructing a blood cell from a gene-transferred hematopoietic cell by using a chromosomally unintegrated viral vector. [MEANS FOR SOLVING PROBLEMS] A blood cell capable of expressing a gene introduced therein can be reconstructed by introducing a negative-strand RNA viral vector into a hematopoietic cell and then performing the hematopoietic cell engraftment using the resulting cell. The gene-transferred hematopoietic cell thus produced can be differentiated into a leukocyte, an erythrocyte and a platelet in the body to which the cell has been engrafted. Thus, there is provided a safe gene transfer system for use in the gene therapy by hematopoietic cell engraftment without causing any damage to the chromosomes of a host.
