PROBLEM TO BE SOLVED: To provide a method for separating a nucleic acid with a substance binding to the nucleic acid at a high rate and releasing the nucleic acid in high efficiency, when pH is raised, by which the nucleic acid can quickly and high efficiently be separated. SOLUTION: This method for separating a nucleic acid from a sample comprises a process for bringing the sample containing the nucleic acid into contact with a two functional groups-having substance having an amino group and a carboxy group and taking a positive charge at the first pH to bind the nucleic acid to the two functional groups-having substance, and a process for releasing the bound nucleic acid at the higher second pH than the first pH, is characterized in that the two functional groups-having substance is a specific substance.
