PROBLEM TO BE SOLVED: To detect a chemical or biological analyte at low cost by a consistent method. SOLUTION: This method 700 of reading an assay comprises a step 702 of illuminating a substrate 100 including a sensitive region 102, and a step 704 of detecting whether the substrate 100 is exposed to an analyte of interest. The step 704 has a step of using a priori data related to low resolution detection 200 and the sensitive region 102 to determine whether the characteristic of a first section of the illumination reflected from the sensitive region 102 is different from that of a second section of the illumination reflected from a substrate 103 out of the sensitive region.
