PROBLEM TO BE SOLVED: To provide a method and an instrument capable of detecting noninvasively an antioxidation state of a biological tissue, and the presence or a risk of malignancy. SOLUTION: This instrument includes a light source 12 for generating light of causing Raman reaction having a wavelength shift to a detected carotenoid, a light emitting and collecting module 14 for emitting the light toward the biological tissue, without damaging the tissue and changing a level of the carotenoid in the tissue by the light, and for collecting scattered light from the tissue, a spectrum selection system 26 for selecting the Raman-shifted light from the collected scattered light, a detection means 28 for scanning and measuring the Raman-shifted light by a frequency characteristic of the carotenoid, and a quantitative determination means 30 for measuring Raman signal intensity with respect to the detected carotenoid. A background fluorescence signal of the biological tissue is removed from a Raman signal of the detected carotenoid, and the presence of abnormality in the tissue is detected based on a substantial difference between the Raman signal intensity and intensity of Raman scattering from the normal tissue.
