The invention relates to a method for determining alleles of the allele group 27 of locus B of the human leukocyte antigen (HLA-B27) from a nucleic acid sample and to a kit for implementing such method. The kit is comprised of allele-specific primers used in the polymerase chain reaction (PCR) in real time, and fluorescently marked allele-specific probes, or an intercalary agent, to detect fluorescence signals. The analysis is completed with the determination of the temperatures of fusion of amplified sequences of nucleic acids. The invention overcomes the limitations of conventional methods for DNA-based typification, in terms of time, possibility of automation, and increase of the resolution, while reducing the number of PCR reactions. It is useful for example to determine the predisposition to a specific disease. |
