Methods for preparing porcine embryo capable of developing into a live born porcine animal comprise obtaining a totipotent porcine nuclear donor cell from a cell culture and forming a cybrid by translocating the cell or nucleus thereof into an enucleated porcine oocyte which prior to enucleation has been matured for between 42 and 56 hours, and which before, during or after translocation the oocytes is activated. The cybrid is then cultured to form a porcine embryo, which can then be transferred into a recipient porcine female to generate a live born porcine animal. Preferably the activation step comprises incubating the cells in a medium comprising ionomycin and/or 6-dimethylaminopurine. (62) Divided out of 512357
