摘要 <P>PROBLEM TO BE SOLVED: To provide a method for designing an optimal siRNA sequence in a target mRNA. <P>SOLUTION: This newly established method for designing an siRNA having optimal activity comprises statistically searching relationships between RNAi activity and characteristics (position, sequence, and base constitution) of the siRNA. It is, namely, found that the siRNA having the optimal activity is designed by statistically searching 702 kinds of siRAN's comprising all of possible designs which target the green fluorescent protein mRNA, so as to develop algorithm, and using the algorithm. Further, activity of an existing siRNA is enhanced by adding an effector sequence which does not require correspondence to a sequence of the target gene and comprises double-stranded 1 to 5 base pairs to a 5' terminal side of an siRNA sequence and/or a 3' terminal side thereof. <P>COPYRIGHT: (C)2005,JPO&NCIPI
