A potassium channel opener having benzofuroindole skeleton and a preparation method thereof are provided, which potassium channel opener stabilizes cell membrane to block or limit excitable cytotoxicity caused by a rapid increase of calcium ion within cells, so that the potassium channel opener is useful for treatment of traumatic or topical anemia or neurodegenerative disorders. The potassium channel opener having benzofuroindole skeleton represented by formula (I) is provided, wherein R1 is hydrogen, R2 is CF3, R3 is COOH, R4 is hydrogen, R5 is hydrogen or chlorine, and R6 is hydrogen or chlorine. The method for preparing the potassium channel opener having benzofuroindole skeleton represented by formula (I) comprises the steps of: reacting a compound of formula (II) with ethylbromoacetate and acetone in methanol solvent containing sulfuric acid to prepare a compound of formula (III); reacting the compound of formula (III) with anhydrous acetic acid and sodium acetic acid to prepare a compound of formula (IV); and reacting the compound of formula (IV) with phenylhydrazin, ethanol, p-TsOH, wherein R1 is 4-CF3 or 4-H, and R2 is 1-COOH, 1-Cl or 4-Cl.
