PROBLEM TO BE SOLVED: To provide a method for activation and amplification of expression of an endogenous gene by homologous recombination.

SOLUTION: Disclosed is a cultured vertebrate cell, wherein a genome comprises an exogenous control sequence, an exogenous exon and a splice-donor site of the 3'-terminal end of an exogenous gene and incorporates a transcription unit comprising a splice-donor site operably linked to the endogenous splice acceptor site of the second endogenous exon of an endogenous gene, the cell includes an amplifiable marker gene derived from constructs, and the genome comprises the exogenous exon as well as all endogenous exon of the endogenous gene.
