A simulated moving layer chromatography separation process wherein a raw material, such as a biomass, containing cellulose and/or hemi-cellulose is hydrolyzed in a hydrolysis apparatus (2) by the use of sulfuric acid into a saccharificated fluid, the saccharificated fluid is subjected to solid-liquid separation in a solid-liquid separation apparatus (3), the resultant filtrate is fed to the simulated moving layer chromatography separation apparatus (5) to separate it into a raffinate and an extract, characterized in that the extract is fractionated into a fraction (A) having a higher sulfuric acid concentration and a fraction (B) having a lower sulfuric acid concentration, the fraction (A) alone is concentrated in a sulfuric acid concentration apparatus (9) and the resulting concentrate is re-used as the sulfuric acid in the above saccharification step. The fraction (B) is re-used as the water for the hydrolysis, water as the eluent for the simulated moving layer chromatography separation apparatus (5) and the like.
