PURPOSE: A method for producing a target substance is provided by using a microorganism which is capable of producing the target substance, and collecting the target substance from the culture medium, thereby reducing the generation of by-products, increasing the production of the target substance and isolating the target substance from the culture medium. CONSTITUTION: The method for producing a target substance comprises the steps of: (a) culturing a microorganism capable of producing the target substance, wherein the system for intake of by-products of the target substance or the substrate of the bio-synthesis system of the target substance is modified in the microorganism; and (b) isolating the target substance accumulated in the cultured medium, wherein the by-products are selected from the intermediate and substrate of the bio-synthesis system of the target substance; the microorganism is Escherichia sp.; the target substance is L-phenylalanine and the by-product is L-tryptophan; the system for intake of by-products is selected from the group consisting of protein Mtr and TnaB; the gene Mtr has the nucleotide sequence of SEQ ID NO:1 or encodes the amino acid sequence of SEQ ID NO:2; and the gene TnaB has the nucleotide sequence of SEQ ID NO:3 or encodes the amino acid sequence of SEQ ID NO:4.
