<P>PROBLEM TO BE SOLVED: To provide a method capable of obtaining a DNA without preparing a conventional gene library and readily synthesizing the DNA from data comprising DNA sequence information. <P>SOLUTION: The subject method for preparing DNA design information on DNA synthesis comprises (1) a step for comparing a partial sequence of a DNA desired to synthesize with an object DNA registered in object DNA database and extracting identical partial sequences (p1-2 and v3-4) and (2) a step for alternately arranging total even number of chains in the order of a sense chain and an antisense chain, starting from a sense chain on 5' end side by using base sequences of template single strands of the extracted partial sequences (p1, p2, v3 and v4) and non-extracted partial sequences (p3-p8), and arranging these chains so that adjacent sense chains and antisense chains and 5' ends and 3' ends have mutually complementary binding regions.
