摘要 PROBLEM TO BE SOLVED: To provide a local polarizing microscope analysis/spectral separation method and an apparatus for the method by composite evanescent wave dark field excitation capable of performing the microspectroscopy of optical isomerism, rotary dependency, circular polarization excitation fluorescence, magnetooptical characteristics, spin dependency, and the like in a minute substance, and to provide a substance operation method and a substance operation apparatus applying the method and the apparatus. SOLUTION: The local polarizing microscope analysis/spectral separation method and apparatus are provided with a means for overlapping two TE (transvers electric field) evanescent waves that are generated on a dielectric surface so that the propagation directions orthogonally cross each other; a means for using a lattice-like local polarization pattern being repeated at a period corresponding to the wavelength of evanescent waves being generated when maintaining it at the same frequency and in a fixed phase difference for lighting or exciting the minute substance 12; a probe 13 having a minute opening 14 for scanning in a two-dimensional direction; integrated, stable polarization analysis systems 15, 16, 17, 18, 19, 20 for selecting the minute substance 12 through the minute opening 14; and a cooling CCD (charge coupled device) 21 for decomposing the polarization of the minute substance 12 by using the polarization analysis systems 15, 16, 17, 18, 19, 20 for measurement.
