PROBLEM TO BE SOLVED: To provide an optical waveguide glucose sensor and a coloring reagent film fixing method for sensitively and accurately analyzing a very minute quantity of glucose in a specimen material 100 such as a body fluid. SOLUTION: The optical waveguide glucose sensor is provided with a substrate 1, a first optical waveguide layer 2 contacting the substrate 1, an entrance grating 3a and an exit grating 3b contacting the first optical waveguide layer 2 and spaced from each other, a second optical waveguide layer 4 contacting the first optical waveguide layer 2, provided between the entrance grating 3a and the exit grating 3b and having a refractive index higher than the first optical waveguide layer 2, a coloring reagent film 8 comprising a monomolecular layer of coloring reagent molecules formed on the second optical waveguide layer 4 and an enzyme catalyst fixing layer 9 containing an enzyme and a catalyst provided and contacting the coloring reagent layer 8.
