PROBLEM TO BE SOLVED: To enable a fluorescent observation and a transmission differential interference observation to easily be switched without causing deterioration in resolution of a sample image nor image deterioration and to enable both at the same time. SOLUTION: A 1/2-wavelength plate 23 is provided on the laser optical path between a laser light source 1 and a scanner unit 7. When a transmission interference observation is made, the polarizing direction of laser light 2 and the optical axis of a lighting-side differential interference element 16 are set at a 45°angle and the lighting-side differential interference element 16 splits the laser light 2 into two light beams 2a and 2b having mutually perpendicular polarizing directions to irradiate the sample S. When a fluorescent observation is made, the optical axis direction h<SB>23</SB>of the 1/2-wavelength plate 23 is slanted by 22.5°to slant the polarizing direction h<SB>3</SB>of the laser light 2 by 45°, and consequently the lighting-side differential interference element 16 neither polarizes nor separates the laser light 2.
