The invention relates to chemistry of heterocyclic compounds, in particular to the process for preparation of 2-amino-3- hetaryl-4(5H)-ketothiophenes of general formula I: I, wherein R=H, CH3, Het = , , , , , , , , , , , , , , , , , which may be used as physiologically active substances or intermediates in synthesis thereof. A process for preparation of 2-amino-3- hetaryl-4(5H)-ketothiophenes of general formula I consists in that 3-(2-hetaryl)-2-keto-3-cyanopropyl ethanthioates of general formula II: II, wherein R = H, CH3, Het as given above, are treated with base in the medium of chemical solvent.
